I have a question about this one. Do you need to stay on the island to get those melchi claims? If so, what is the best place for them?

No, you can do it anywhere, even on F.O.M.A (which counts as Calypso area).
I got my Melchi's near the Orthos oil field (where the new oilrig is, North of Camp Icarus).
